Leukocyte changes during acute cross-circulation experiments between leukemic and normal or irradiated rats.

چکیده
TVitIi the technical assistance of MARY CHALTAS T HI’ viability amid fate of circulating and tramisfused mammalian leukocytes are poorly understood. In order to investigate sonic of these problems more completely high leukocyte coumit (leukemic) domior rat-s were cross-circulated with normal and leukopemiic recipiemits. Post cross-circulatiomi hematologic observations its these animals imidicate that donor-recipient leukocyte concentratiomi relatiomiships may be an important determinant of blood leukocyte survival. Leukopenic rats retaims beukocytes in their peripheral blood for very short periods of time folbowimig cross-circulation with msormal animals.’ There is good evidemice, however, thatthe cross-transfused beukocytes remain viable its mioncirculatitig sites for much bomiger periods of time.’ 2 These data suggest that the blood survival time of traissfused leukocyt-es might be imicreased if the “pool” of tissue leukocytes was normally saturated, amid acted to impede circulatory loss. Its order to test this hypothesis attempt-s first were made to “saturate” leukopetsic animals with large volumes of leukocytes in order to slow the rate of peripheral leukocyte removal. Secondly, the peripheral leukocyt-e counts of normisl rats were elevated greatly through cross-circulation with leukemic rats in order to demonstrate the ability of normally saturated reserves to affect more prolomiged circulation of the transfused cells.

IN PREVIOUS STUI)IES imivolvimsg acute cross-circulat-ioms experiment-s in rats, evidence was obtained that leukocyt-es transfused by this method were not imnsediately destroyed. ‘ 2 It was postulated that disappearance of t-ramisfused leukocytes fromn the arterial blood of irradiated leukopenic animals represemsted redistribut-ioms of these cells into a depleted body leukocyte “pool,” rather t...
متن کاملA qualitative study of normal leukocyte antigens of human leukemic leukoblasts.
Twelve leukocyte antigens have been determined by the method of cytotoxicity in v~tro on leukoblasts from 38 patients with acute leukemia, using two batteries of isoimmune human sera. None of these antigens was absent in this leukemic population. The presence of the leukocyte antigens 1, 3, 4, 6, and 7 of the Hu-1 system (5) was confirmed by absorption experiments. In 17 patients, the leukocyte...

Kinetic studies in cultures containing 2 X 10(5) peripheral leukocytes from patients with acute myelobastic leukemia revealed extensive, radiation-sensitive increases in thymidine incorporation without parallel increases in cell number. Modest and variable stimulation of 3HTdR incorporation was seen with the addition of either leukocyte-conditioned medium prepared with phytohemagglutin (PHA) or...
